hplc測(cè)定復(fù)方厚樸顆粒中厚樸酚及和厚樸酚的含量
復(fù)方厚樸顆粒;,,高效液相色譜法;,,厚樸酚;,,和厚樸酚 摘要:目的建立hplc法測(cè)定復(fù)方厚樸顆粒中厚樸酚及和厚樸酚的含量。方法采用diamondsilcmc18色譜柱(6.4 mm×200 mm,5 μm);流動(dòng)相為甲醇-0.1%磷酸溶液(78∶22);流速1 ml/min;柱溫35℃;檢測(cè)波長(zhǎng)294 nm。結(jié)果厚樸酚的線性范圍為0.041 1~0.822 2 μg,r=0.999 9,平均回收率為97.1%,rsd=1.27%;和厚樸酚的線性范圍為0.023 06~0.461 2 μg ,r=1,平均回收率為96.7%,rsd=1.25%。結(jié)論該方法結(jié)果準(zhǔn)確,重現(xiàn)性好,可作為復(fù)方厚樸顆粒質(zhì)量控制的定量方法。 關(guān)鍵詞:復(fù)方厚樸顆粒; 高效液相色譜法; 厚樸酚; 和厚樸酚 上禾生物 determination of magnolol and honokiol in compound houpu granule by hplc xia hou lin ,sheng yan,zhou xiao mei,wu xi,dong min,tan ling (chengdu university of traditional chinese medicine, 610072 ,china) abstract:objectiveto establish an hplc method for the determination of magnolol and honokiol in compound houpu granule.methodsthe samples were analyzed on a diamondsilcmc18 column (6.4 mm×200 mm,5μm). the mobile phase was a mixture of meoh-0.1%h3po4(78:22);the rate of flow was 1ml/min; the column temperature was 35 ℃; the detection wavelength was 294 nm.resultscalibration graph was linear in the range of 0.041 1~0.822 2 μg,r=0.999 9 for magnolol, 0.023 06~0.456 12μg, r=1 for honokiol. the recovery was 97.1%, rsd=1.27% for magnolol; 96.7%,rsd=1.25% for honokiol.conclusionthe method is sensitive and its results are accurate with good reproducibility .it can be used for the quality control of compound houpu granule. key words:compound houpu granule; hplc; magnolol; honokiol 復(fù)方厚樸顆粒由厚樸、藿香、蒼術(shù)等藥物組成,具有解表和胃之功效,用于感受風(fēng)寒,內(nèi)有食積,寒熱吐瀉等癥。為了控制本品質(zhì)量,保障用藥安全與有效,本文采用高效液相色譜法測(cè)定方中君藥厚樸的主要成分厚樸酚、和厚樸酚的含量[1,2]。 1 儀器與試劑 agilent 1100高效液相色譜儀(美國(guó)agilent);agilent 1100二極管陣列檢測(cè)器美國(guó)(agilent 1100);agilent 1100高效液相色譜工作站(美國(guó)agilent);厚樸酚、和厚樸酚對(duì)照品(中國(guó)藥品生物制品檢定所,供含量測(cè)定用,批號(hào)分別為110729 200208、0730 200206);復(fù)方厚樸顆粒(自制);甲醇為色譜純(fisher),其它試劑均為分析純。 2 方法與結(jié)果 2.1 色譜條件diamondsilcmc18色譜柱(6.4 mm×200 mm,5μm迪馬公司);流動(dòng)相甲醇 0.1%磷酸(78∶22);流速1 ml/min;柱溫35℃;檢測(cè)波長(zhǎng)294 nm;理論板數(shù)以厚樸酚峰計(jì)算不少于3800。在此條件下厚樸酚與和厚樸酚分離良好。
