建立測(cè)定胃康膠囊中厚樸酚、和厚樸酚含量的高效液相色譜法。
方法 色譜柱為hypersil ods c18 (4.6 mm×100 mm,5 μm)；流動(dòng)相為甲醇-水(70∶30),檢測(cè)波長(zhǎng)為294 nm；柱溫為30 ℃；流速為1.0 ml/min。 結(jié)果 厚樸酚在0.053 4～1.068 μg范圍內(nèi)線性關(guān)系良好(r=0.999 8),平均回收率99.03%,rsd為0.68%；和厚樸酚在0.09～1.80 μg范圍內(nèi)線性關(guān)系良好(r=0.999 8),平均回收率為98.95%,rsd為0.69%。 結(jié)論 本方法簡(jiǎn)便、快速,測(cè)定結(jié)果準(zhǔn)確,重現(xiàn)性好�？勺鳛樵撝苿┑馁|(zhì)量控制指標(biāo)。 【關(guān)鍵詞】 胃康膠囊；厚樸酚；和厚樸酚；高效液相色譜法；含量測(cè)定 determination of the contents of magnolol and honokiol in weikang capsule by hplc peng ping-jian huaihua first peoples hospital of hunan province, huaihua 418000, china abstract：objective to establish a hplc method for determining the contents of magnolol and honokiol in weikang capsule. methods stationary phase was hypersil ods c18 (4.6 mm×100 mm, 5 μm), mobile phase was methanol-water (70∶30), with detection wavelength 294 nm, flow rate 1.0 ml/min and column temperature 30 ℃. results there was good linearity in range of 0.0534～1.068 μg (r=0.999 8, n=6) for magnolol. the average recoveries of magnolol was 99.08%, rsd was 0.57%. there was good linearity in range of 0.09～1.80 μg (r=0.999 8,n=6) for honokiol. the average recoveries of honokiol was 98.95%, rsd was 0.69%. conclusion this method is simple, fast and accurate for the quality control of the preparation. key words：weikang capsule；magnolol；honokiol；hplc；content determination 胃康膠囊為我院內(nèi)科傳統(tǒng)制劑,功效健脾和胃、行氣止痛。主治淺表性、糜爛性、萎縮性胃炎。用于惡心、嘔吐、泛酸、燒心、食欲不振、胃脘部疼痛、胃部怕冷、喜暖、喜按等癥。該制劑由黨參、厚樸、白芍、砂仁等多味中藥組成。其中厚樸為君藥,具有燥濕消痰、下氣除滿的功能[1],在方中起重要作用。厚樸的主要成分為厚樸酚、和厚樸酚等[2],含量較高,易控制,可選為定量指標(biāo)。本實(shí)驗(yàn)采用hplc法測(cè)定胃康膠囊中厚樸酚、和厚樸酚的含量。 1 儀器與試藥 高效液相色譜儀：agilent1100系列四元梯度泵、agilent1100手動(dòng)進(jìn)樣器、agilent1100系列二極管陣列檢測(cè)器、agilent1100工作站(agilent公司)。超聲波清洗器(昆山市淀山湖檢測(cè)儀器廠),萬(wàn)分之一電子分析天平(沈陽(yáng)龍騰電子稱量?jī)x器有限公司)。 厚樸酚(中國(guó)藥品生物制品檢定所,批號(hào)0729-200309),和厚樸酚(中國(guó)藥品生物制品檢定所,批號(hào)0730-200206)。胃康膠囊(本院自制,批號(hào)050613、051110、060306)。 甲醇(色譜純,上海陸都化學(xué)試劑廠,20050623),其余所用試劑均為分析純。 2 厚樸酚與和厚樸酚的含量測(cè)定 2.1 色譜條件 色譜柱：hypersil ods c18(4.6 mm×100 mm,5 μm)；流動(dòng)相：甲醇-水(70∶30)；檢測(cè)波長(zhǎng)：294 nm；柱溫：30 ℃；流速：1.0 ml/min；理論塔板數(shù)以厚樸酚峰計(jì)不低于3 800
